Hold "control:Ctrl" + roll up your Mouse wheel to larger the icons on your Desktop.

hard to tell, because dock icons usually arent displayed in 256x256. create a proper set in 128, 64 and 32 resolutions, then we can judge if it works

Good idea. I'm using HD icons because I prefer bigger dock icons. But some more sets will be reasonable.
